Barbara Randolph war die Tochter der Schauspielerin Lillian Randolph (1898-1980).

Mary Wilson behauptet in ihrem Buch Dream girl, dass eine gewisse Barbara Randolph und Betty Kendrick Hintergrundsängerinnen auf der einzigen Single Tears of sorrow (1960) der Primettes gewesen seien; allerdings ist nicht bekannt, ob es sich dabei um dieselbe Barbara Randolph handelt. Die Primettes wurden 1961 in Supremes umbenannt.

1964 veröffentlichte Randolph auf dem Plattenalbel Mercury ihre erste eigene Single, Sweet daddy tree top tall, die allerdings kein Erfolg wurde. Nach dem Ausstieg von Zola Taylor war Randolph ab 1964 Sängerin bei den Platters und sang auf deren letztem Album für Mercury, The new soul of the Platters (Campus style) (1965), bei fünf Stücken die Hauptstimme (darunter das vom Manager der Gruppe, Buck Ram, geschriebene "The big forget"). 1965 wurde sie durch Betty Jackson ersetzt, die ihrerseits 1966 durch Sandra Dawns ersetzt wurde.

Angeblich soll Randolph 1967 als möglicher Ersatz für die bei den Supremes ausgestiegene Florence Ballard im Gespräch gewesen sein. Diana Ross soll jedoch das gute Aussehen von Randolph missfallen haben, sodass nichts daraus wurde. Stattdessen veröffentlichte Randolph bei Motowns Sublabel Soul ihre zweite Single I got a feeling; das Lied war von Holland-Dozier-Holland als B-Seite der Four Tops-Single Bernadette (1967) geschrieben worden.

Als Tammi Terrell krank wurde, ersetzte Randolph diese auf einer kurzen Tournee mit Marvin Gaye. Kurze Zeit darauf veröffentlichte sie ihre dritte Single, Can I get a witness (1963), eine Neufassung des Marvin Gaye-Klassikers von 1963. Nachdem auch diese Single kein Erfolg wurde, war ihre Karriere bei Motown bereits wieder beendet. 15 weitere, 1969 für Motown aufgenommene Titel blieben unverständlicherweise bis 2003 unveröffentlicht. Ausserdem existiert auch noch ein Alternativ-Take von "When it rains it pours".

Später heiratete Randolph den Komponisten Eddie Singleton (geboren 1931), der zuvor bereits mit Raynoma Gordy (Berry Gordy's zweite Frau) verheiratet gewesen war. Randolph und Singleton gründeten dann zusammen eine Produktionsfirma.

Nach einigen wenig beachteten weiteren Singles nahm sie 1989 eine Neufassung von "I got a feeling" für das britische Plattenlabel Nightmare und einige Jahre später für Motorcity das Album Breaking into your heart (1992) auf.

Barbara Randolph starb 60-jährig am 15. Juli 2002 in Südafrika an Krebs. Etwa 2008 starb auch ihr Mann Eddie Singleton.
